Production of Non - dormant Bulblets of Lilium Oriental Hybrid by Control of Culture Temperature and Growth Regulators In Vitro

Abstract
This study was performed to investigate dormant status of bulblet regenerated from microscale in vitro and to produce non-dormant bulblet in Lilium Oriental Hybrids. All bulblets regenerated under the dark condition had dormant irrespective of cultivars, culture periods and sucrose concentrations. But, culture temperature lower than 15¡É and addition of 1.0 §·¡¤L^(-1) BA or 1.0 §·¡¤L^(-1) fluridone in the culture medium was effective in producing non-dormant bulblets. Especially, addition of fluridone, an inhibitor of ABA-synthesis, in the medium was effective to produce non-dormant bulblets irrespective of cultivars and culture conditions. Morphological and physiological differences were appeared by fluridone, especially leaf elongation was stimulated in high concentration. The specific and total activities of peroxidase of non-dormant bulblets regenerated in vitro were higher than those of dormant bulblets, but protein content was same in the bulblets. Peroxidase isozymes of non-dormant bulblets were greater in number and amount than those of dormant bulblets.
